medical cause of depression in NEETs and others

The reasons are startlingly simple.
1. Self-actualization
The NEET is unable or unwilling to translate his desires and objectives into the real world. This is not required to be a NEET, it is just what I have observed from countless NEETs.
2. Not active in society
We are social animals, and we are evolved to function as a group. When a member is excluded or voluntarily leaves, he is not contributing or participating in society or human development. This naturally makes the NEET sad.
3. Other basic needs
Due to the above two reasons, the NEET often does not meet many basic needs. He does not eat healthy, exercise, or recieve any social interaction. A NEET with former trauma will never supplant those memories with new ones.

In conclusion, NEETs are sad because they do not meet basic needs, which exist in order to further human development.

          Then these "neets" are those which only technically fit the definition of NEET but are not the kind of people you think of when you think NEET. They just happen to be unemployed and don't have any deep social phobias. Instead of calling those people "NEETs", you might want to simply call them "unemployed" or "in between jobs" or "early retirees"
