Literally the entire definition of 'manga' is that it's a comic and it's from Japan. If it meets those qualifications it's manga by definition.

Definitions are man-made. "Video game" includes mobile and browser games, which obviously ought to be excluded in discussing people who play video games. "Books" includes YA and chick lit which obviously ought to be excluded when discussing serious readers. That's the whole premise of the distinction "real" / "not real." It's a further subdivision of a categorization that is too broad for some purpose. In this case, the purpose was determining which gender reads more manga. If it turns out that there's an entire subset of comics from Japan that any serious weeb would scoff at and call "not real manga," then the extra categorization is useful.
